What I'm mostly concerned about now is the stress level. As of now, my gecko isn't one to be stressed easily (handles well, eats easily, doesn't even mind being - supervised! - around my cat), but I'm worried that a car trip will make him unsettled. He won't be rolling around or anything, but is this a huge concern with anyone?

Have you been able to find a picnic-style styro container and some reptile heating packs for your return trip to school? (The heating packs can be ordered online.) Get a deli cup or a medium Kritter Keeper not all that much bigger than he is to contain him. Poke holes in that so he can breathe. Scrunch up a couple paper towels to cushion him and place them in the deli cup/Kritter Keeper. Add you gecko. Then that container goes into the styro box. No need to use a heat pack unless you encounter a temperature emergency.
(Don't place the paper towels, then the gecko, directly into the styro picnic container without the deli-cup/Kritter Keeper first.)
I would also bring a spray bottle for gecko water in case of some emergency.
I think he will be just fine. How does that sound?
